Now that I've read the fic, I agree with you about the snippet given that it comes directly after Dean specifically brings up the cheek kissing and hand-holding in a negative way. Without that, I could have assumed that Dean was just indicating that he was uncomfortable being thought of as gay. But bringing up the cheek kissing and hand-holding is just weirdly assuming a sexual context to them. Siblings kiss each other on the cheek and hold hands in real life (though, yes, brothers don't usually walk around holding hands), but Dean very much tossed it out like a sexual accusation.
